Percutaneous absorption of zinc from zinc oxide applied topically to intact skin in man

Ågren measured zinc from a 25% zinc oxide occlusive dressing on intact adult forearm skin for 48 hours.

Ågren measured zinc from a 25% zinc oxide occlusive dressing on intact adult forearm skin for 48 hours. The text layer of this PDF is empty apart from download stamps. Tables were read from the page image. Table 1 means are 2.91 mg/cm2 in the new dressing and 2.68 mg/cm2 after 48 hours (n=10). The results text gives the difference as 0.23 mg/cm2 (95% confidence limits 0.05 to 0.40) and an average flux of 5 µg/cm2/h. Table 2 means for epidermis are 35 µg/g dry weight on control skin and 187 µg/g under the dressing. Blister fluid means are 0.27 and 0.95 µg/ml. In vitro, dressing zinc fell by 0.54 ± 0.01 mg/cm2 at pH 5.4 and by 0.04 ± 0.04 mg/cm2 at pH 7.4 over 48 hours. The print year is 1990 (Dermatologica volume 180, pages 36-39). Crossref records DOI 10.1159/000247982 with an issued date of 2009-10-08, which is the DOI registration, and the same page span.

Methods (brief)

Suction blisters at 250 mm Hg. Epidermis and dermis dried 24 h at 105 C and ashed at 450 C. Flame AAS. In vitro release through a cellulose acetate membrane into diluted horse serum at pH 5.4 and 7.4.

Detection and reporting: Not stated. Reference material 125 µg/g versus certified 123 ± 8 µg/g. Adhesive duplicate CV 1.4%.

Basis: Dressing zinc in mg/cm2. Epidermis dry weight. Blister fluid per ml.

Evidence fitness

This source can support: zinc delivery from a 25% zinc oxide occlusive dressing into adult epidermis and blister fluid; pH dependence of zinc release from that dressing in vitro.

It cannot support: infant skin; diaper-cream vehicles; lead or cadmium in zinc oxide; a regulatory limit.

Limitations

The PDF text layer does not contain the article. Table values were read from the page image. A stripping experiment in one subject is described in the OCR text and is not used here because that sentence was not re-read from a crop. This dressing study is not a diaper-cream trial. It is a different paper from Ågren 1991 in Acta Dermato-Venereologica.
