Laboratory specimens illustrating Wiester 1975 palladium research context.

Cardiovascular actions of palladium compounds in the unanesthetized rat.

Overview

This study characterized cardiovascular effects of intravenous palladium compounds in unanesthetized surgically prepared rats, monitoring ECG, aortic blood pressure, cardiac contractility (dp/dt), and respiration. The minimum effective IV dose for cardiac arrhythmias was 0.4 mg Pd/kg as PdSO4. The paper also notes historical use of palladium hydroxide as a subcutaneous injection for obesity treatment, which caused weight loss and fever.

Key numbers

  • 41 rats given 0.25-2 mg/kg Pd ion IV
  • Minimum effective IV dose for cardiac arrhythmias: 0.4 mg/kg Pd as PdSO4
  • Mild effects mean dose: 0.4 +/- 0.2 mg/kg as PdSO4
  • PdSO4, Pd(NO3)2, and PdCl2 showed similar cardiovascular activity
  • (NH4)2PdCl4 required 3x higher Pd dose to achieve equivalent cardiovascular response
  • Historical Pd(OH)2 subcutaneous treatment (obesity): 5-7 mg/day for 3 months caused weight loss and fever
  • All findings from parenteral administration only

Methods (brief)

Unanesthetized surgically prepared rats instrumented for ECG, aortic blood pressure, cardiac contractility (dp/dt), and respiratory monitoring. IV administration of palladium salts at graded doses. 41 rats total across dose groups. Oral and inhalation routes not addressed.

Implications

This page makes the source discoverable for palladium evidence routing. Provides cardiovascular toxicity thresholds for IV palladium relevant to understanding systemic effects from parenteral exposure. All toxicity demonstrated via parenteral routes; dietary exposure implications require extrapolation given the low oral bioavailability demonstrated by Moore et al. 1975.
