Nickel, palladium and rhodium induced IFN-gamma and IL-10 production as assessed by in vitro ELISpot-analysis in contact dermatitis patients

Overview

This study used ELISpot assays to characterize IFN-gamma and IL-10 producing peripheral blood mononuclear cells (PBMCs) from contact dermatitis patients stimulated with nickel, palladium, and rhodium salts in vitro. The work identified optimal stimulation concentrations for each metal, determined toxicity thresholds, and characterized how IL-10 co-production modulates the Th1 (IFN-gamma) response in metal-sensitized patients.

Key numbers

  • Optimal Pd stimulation concentration: 2.5 ug/mL PdCl2 (highest IFN-gamma ELISpot frequency)
  • Pd cytotoxicity threshold: greater than 25 ug/mL (inhibits PHA-induced cytokine production by >25%)
  • Optimal nickel (Ni) stimulation: 20 ug/mL NiSO4
  • Ni cytotoxicity threshold: greater than 50 ug/mL
  • Rh optimal stimulation: 5 ug/mL; cytotoxicity threshold: greater than 30 ug/mL
  • Pd-ACD patient group n=2; Ni-ACD patient group n=3
  • ELISpot indices expressed as metal-stimulated vs PHA-stimulated ratio

Methods (brief)

PBMC isolation from venous blood of contact dermatitis patients (confirmed by patch test). ELISpot assay for IFN-gamma and IL-10 cytokine-producing cells after 48h stimulation with graded metal salt concentrations. Toxicity threshold defined as the concentration inhibiting PHA-induced cytokine production by more than 25%. Optimal concentration identified as the stimulation level producing maximum ELISpot frequency.

Implications

This page makes the source discoverable for palladium evidence routing. Provides optimal in vitro stimulation concentrations for Pd in immunological assays and characterizes the cytokine response profile (IFN-gamma/IL-10 balance) in Pd-sensitized patients, relevant for understanding immune mechanisms and designing diagnostic assays.
